Denton signed off after six seasons and nearly 200 episodes with final interviews with Hollywood actor and director Ben Stiller and Wendy Whiteley.
"It`s been a privilege and education to talk to such an extraordinary range of people," Denton said before the credits rolled.
"We are grateful to so many guests who`ve allowed us to hold their hearts and souls up to the light."
Enough Rope has been a ratings drawcard for the ABC since March 2003, with major highlights being interviews with former US President Bill Clinton, disgraced AFL star Wayne Carey, Steve Irwin, Paul Keating, Kerry Packer and Heath Ledger.
The final show last night came just days after the program won its fourth AFI trophy.
It was named best light entertainment television series at the Australian Film Institute Industry Awards in Melbourne.
